Measurement of SIRT1 by Western blotting following transient transfection with plasmid pCMV6-ENTRY-SIRT1 (Origene) or with vector control (b) or with siRNA targeted to SIRT1 or with control siRNA (c). Approximately 10 μg of protein was loaded in each lane. Three biological replicates are presented for each condition. Approximate molecular weights are indicated. “+SIRT1” indicates cells transfected with pCMV6-ENTRY-SIRT1; “control” indicates cells tranfected with vector control; “siRNA” indicates cells transfected with one of two siRNAs (#1 or #2) targeted to SIRT1 or with a control siRNA.
